rabiei, the causal agent of Ascochyta blight of chickpea (Cicer arietinum L.), and toxin production-by the wild type

چکیده
Ascochyta rabiei is the causal agent of blight of chickpea, an important food legume crop for human populations in Developing Countries. All reliably identified isolates of the fungus produce toxins in culture, known as the solanapyrones, of which solanapyrone A is the most frequently found and also the most toxic. The principal aim of the project was to determine the role of this toxin in the disease syndrome by producing toxin-minus mutants and testing them for virulence. Four transformation techniques were attempted: Restriction Enzyme Mediated Integration (REMI), electroporation, particle bombardment and Agrobacterium tumefaciens-mediatod transformation. With the last, employing a T-DNA containing a hygromycin resistance gene, 908 transformants were obtained from germinated pycnidiospores on a selective medium containing hygromycin. Genuine transformants were tested for the production of solanapyrone A using an assay in microtitre plates. Loss of toxin production by transformants was confirmed by reversed phase High Performance Liquid Chromatography. Sixteen transformants produced significantly less solanapyrone A than the wild-type strain. Transformants were also screened for integration events by PCR, using primers specific to the hygromycin resistance gene and homologous hybridisation to a probe consisting of this gene. Among the four transformants tested, three have integrated two copies of T-DNA and one had a single insertion. In order to optimise the production of solanapyrone A so as to provide a source of the compound for screening chickpea genotypes, three types of cultures of A. rabiei were tested: still culture, shake culture and fermenter culture. The toxin was purified from culture filtrates by solvent partitioning followed by flash chromatography. The effect of two safeners on the sensitivity of chickpea shoots to solanapyrone A was tested using bioassays. Dichlormid (300 or 800 jig per shoot) and fenclorim (18 pg per shoot) decreased the sensitivity of chickpea shoots to solanapyrone A 1.6 and 2.5-fold, respectively.

Isolation and Characterization of Rhizosphere Bacteria for the Biocontrol of the Asochyta Rabiei in Iran
Chickpea (Cicer arietinum), a cool season grain legume grown on a surface of 11 million ha with worldwide production of about 9 million tons [1], is cultivated in more than 45 countries throughout the world including Iran where it is grown mostly in rainfed areas and on marginal lands in the province of West Azarbayjan. Ascochyta blight is perhaps the most frequent and damaging disease of chick...

ABSTRACT Historically, the response of chickpea (Cicer arietinum L.) to Didymella rabiei (causal agent of Ascochyta blight) has been mainly related to as complete resistance and it was commonly assayed with qualitative (nonparametric) scales. Two reciprocal populations, derived from intra-specific crosses between a moderately resistant late flowering Israeli cultivar and a highly susceptible ea...
متن کاملSympatric ascochyta complex of wild Cicer judaicum and domesticated chickpea
The aim of this study was to isolate, identify and characterize ascochyta blight pathogens from Cicer judaicum , a wild annual Cicer species which grows in Israel and other Mediterranean countries in sympatric distribution with legume crops, and determine their virulence and aggressiveness to other wild and domesticated legumes. Native C. judaicum plants exhibited symptoms resembling ascochyta ...

Fungi colonizing senescent chickpea (Cicer arietinum) stems and postharvest debris from Pullman, WA, were enumerated and identified with the objective of finding species potentially useful for biological control of Didymella rabiei (conidial state 1⁄4 Ascochyta rabiei), causal agent of Ascochyta blight. In addition to D. rabiei, primary colonizers were, in order of decreasing abundance, Alterna...
متن کاملDifferent ecological affinities and aggressiveness patterns among Didymella rabiei isolates from sympatric domesticated chickpea and wild Cicer judaicum.
Domesticated chickpea (Cicer arietinum) and its wild relative C. judaicum grow in sympatric distribution in Israel and both are susceptible to Ascochyta blight caused by Didymella rabiei. C. arietinum was grown for millennia in drier and hotter Levantine spring conditions while C. judaicum grows in the wetter and milder winters. Accordingly, it is possible that D. rabiei isolates originated fro...
